I wouldn't say he's the best guest ever, but I do remember not expecting anything from an interview with this guy and being shocked at how cool he was. Honorable mention, at least... Paul-Fuckin'-Anka.
Brad Garrett was also my first thought, the last great guest before I quit listening.
It was one of the very first Sirius interviews, but I gotta go with Gianni "Carlo" Russo from the Godfather. That was a phenomenal interview, having very little to do with Howard.

I would like to amend my question. No one shot guests allowed (ie, Brad Garrett)
C'mon! The easy answer is David Arquette. His 14th guest appearance for the year 2011 was especially outstanding. I remember when Howard asked him about his penis size and he began giggling like a pedophile at a children's beauty pagent. It was pure comedy gold.
I guess I'm the exception, but I always got a little bummed when they announced a guest was coming in. Any guest. Didn't matter who. The show was always more entertaining when it was just the gang riffin' or busting each others balls. When I say gang, I mean Howard, Robin, Fred, Jackie, Gary and Stuttering John.
There are no balls left to be busted, bud.
